一、產品介紹

Genscript 提供穩定表現 GPCR 或免疫相關蛋白的細胞株。GPCR (G protein-coupled receptors) 為常見的訊息傳遞路徑的受體,參與許多功能,包括神經、肌肉和生長等等,因此很多藥物是透過結合 GPCR (抑制或活化) 來達到療效。為了大量篩選藥物,必須先建構一個穩定表現 GPCR 的細胞株。因此,Genscript 建構的細胞株可以大量節省時間和人力。
Genscript 提供的免疫相關蛋白表現的細胞株包含三類,第一類是 Immune checkpoint for binding assay (免疫抑制蛋白),細胞表現免疫抑制的蛋白,用來針對癌症免疫療法的研究和藥物篩選。第二類是 Immune activation for functional assay (免疫活化蛋白),細胞表現免疫活化的蛋白,給予相對應的 ligand 可以活化細胞,促使 IL-8 的分泌。第三類是 Fc Receptor,細胞表現 Fc receptor,可以結合抗體的 Fc region,可以用來研究抗體的結合 (binding assay)。三類不同的細胞株可以應用在不同層面,可以拿來測試結合力,也可以拿來做篩選藥物的平臺。Genscript 提供的這些細胞株都能夠為實驗者省下大量時間和人力。

二、產品特色

GenScript 提供的特殊蛋白表現細胞株 (兩管,1x106 細胞/管):

1. 皆經過 FACS 確認蛋白穩定表現:
經過超過15代以上培養,以 FACS 確認,因此確認蛋白質穩定表現在細胞膜上。
以 FACS 分析 CHOK1 (紅) 和 CHO-K1/PD-1 (藍) 細胞的 PD-1 表現量。 以 FACS 分析 GS-H2 (紅) 和 GS-H2/CD27 (藍) 細胞的 CD27 表現量。
 

2. 節省篩選藥物前置作業時間和人力:
篩選藥物前必須先建構穩定表現蛋白的細胞株,因此 Genscript 建構的細胞株可以作為篩選藥物平臺,省下大量細胞建構時間和人力。

3. GPCR 表現細胞株和免疫活化蛋白表現細胞株經過功能性確認:
GPCR 表現細胞株給予相對應的 ligand 後,路徑皆能夠活化,促使鈣離子的流動。
免疫活化蛋白表現細胞株給予相對應 ligand 後,皆能夠使免疫細胞活化,促使 IL-8 的分泌。
CHO-K1/M1 和 CHO-K1 細胞給予 M1 receptor 活化物 carbachol,並以 calcium-4 螢光染劑偵測鈣離子,透過 FlexStation 儀器紀錄。以相關螢光數值 [relative fluorescent units (RFU)] 對 log [carbachol 濃度] 做圖。 功能性分析確認,給予 GS-H2/CD27 細胞 CD70,檢測活化後產生的 IL-8。
